Extraction technology of forsythiaside from fructus forsythiae by the method of orthogonal test optimization and separation through macroporous resin
【Abstract】 Objective To design the extraction and separation technologcial conditions of forsythiaside from Fructus Forsythiae. Methods The solvent of extraction was investigated. The orthogonal test method was adopted to investigate the effect of the 4 factors, which include the concentration and volumn of alcohol, the time and the times of extraction and the process of absorption separation through macroporous resin column on the yielding rate of forsythiaside from Fructus Forsythiae. Results The optimum extraction condition were: extracting 3 times with 70% alcohol, 1.5 h each time with 7 times the volume of the alcohol. The eluent was 40% alcohol. Conclusion The results indicate that the yielding rate of forsythiaside from Fructus Forsythiae was from 9.1% to 10.6% by this technology. The concontration of forsythiaside in extract was from 18% to 22%, so this technology is suitable for industrial production.

连翘为木犀科植物连翘Forsythia suspensa (Thunb.)vahl 的干燥果实。连翘中所含成分主要为连翘酯苷(forsythiaside)[1, 2]、连翘苷(phillyrin)[3]、挥发油[4]、黄酮类等。其中,含量较高且药理活性较强的成分是连翘酯苷,其对金黄色葡萄球菌等11种细菌有极强的抑制作用,是连翘抗菌、抗病毒的主要有效成分之一[5] 。故本文以连翘酯苷含量为指标,考察了提取溶媒。确定提取溶媒后,采用正交试验进行提取工艺优化,提取物进一步用大孔树脂分离纯化,从而系统的研究了连翘提取和分离纯化工艺。
1 仪器与试药
Waters高效液相色谱仪(616泵,996二极管阵列检测器;717自动进样器,2010色谱管理系统);甲醇(色谱纯,国药集团化学试剂有限公司);水(重蒸馏水,自制);磷酸(分析纯,国药集团化学试剂有限公司);连翘酯苷对照品(沈阳药科大学徐绥绪教授提供)。
2 方法与结果
2.1 提取溶剂的选择 连翘中含量最高抗菌活性最大的成分为连翘酯苷,其结构中含双糖基和咖啡酰基,极性较大,在水和含水乙醇中溶解性极好。因此分别选择以水为溶媒的煎煮法和以70%乙醇为溶媒的回流提取法,比较二者优劣,含量测定结果见表1 表1 不同溶剂提取的结果结果表明:70%乙醇对连翘酯苷的提取率较水高,故选择醇提取法。
2.2 醇提取工艺优化的研究 采用正交试验法对乙醇提取工艺中的加醇量、回流时间、回流次数、醇浓度进行考察,方法与结果如下。
